Step 1: Std. Calibrating curve


10 mg drug was dissolved in pH 7.4 Phosphate Buffer, to prepared 100 ug/ml solution

0.2 ml solution is diluted with 10 ml Buffer

2 g/ml solution

0.4 ml solution is diluted with 10 ml Buffer

4 g/ml solution

0.6 ml solution is diluted with 10 ml Buffer

6 g/ml solution

0.8 ml solution is diluted with 10 ml Buffer

8 g/ml solution

1.0 ml solution is diluted with 10 ml Buffer

10 g/ml solution

1.2 ml solution is diluted with 10 ml Buffer

12 g/ml solution

(Above all solutions are 10 g/ml solution)

Step 2: Take absorbance of all solution and Plot graph in MS Excel.

Step 3: After taking absorbance, to plot graph in Excel, Click Insert

Abs.: Taking solution after suitable time interval, to take absorbance of this solution, if absorbance is
more than 1, to dilute the solution.

Conc g/ml: 1. To calculate Conc g/ml with the help of Y = mx + c (Equation: std.
calibrating curve) this equation,
2. To convert this equation in x = Y-c / m (Y = Abs.)
DF: dilution factor is indicate number time to dilute the given solution (10, 100 and 1000).

Conc g/ml: DF * Conc g/ml to get Conc g/ml.


Conc mg/ml: Conc g/ml / 1000 to get Conc mg/ml.
Conc (mg/0.3ml): Conc mg/ml * 0.3 to get Conc mg/0.3ml.
Conc mg/10ml: Conc mg/ml * 10 to get Conc mg/10ml.
DR (drug release): to select reading in below the zero in DR column * First reading 0 in
Conc mg/10ml to get DR.
%Diff: (% Diffusion rate): (DR*100) / Labelled Claim of drug to get %Diff.

IMPORTANT NOTE:
1. In every calculation in Excel sheet, Always starting the calculation with
(equal to sine), if youre not used this sine calculation was not done.
