Methodology Preparation of Solutions: HMR MR

This document outlines the methodology for preparing various solutions used in an experiment to determine the acid dissociation constant of methyl red. Solutions of methyl red, sodium acetate, acetic acid, and hydrochloric acid were prepared in different concentrations. Ten sample solutions were also prepared with varying quantities of the components. The absorbance spectra of the methyl red solutions were analyzed to determine the wavelength maxima for the acidic and basic forms. Absorbance readings of the samples at these wavelengths were then used to calculate the molar absorptivity constants and acid dissociation constant of methyl red.

METHODOLOGY

Preparation of Solutions
The following solutions were prepared prior to the experiment proper: 50 mL of methyl red stock
solution, 50 mL of methyl red standard solution, 100 mL of 0.040 M NaOAc from NaOAc3H2O, 50
mL of 0.010 M NaOAc from 0.040 NaOAc, 25 mL of 1.0 M HOAc, 50 mL of 0.020 M HOAc from
1.0 M HOAc, 50 mL of 0.10 M HCl, 50 mL of 0.010 M HCl from 0.10 M HCl, and 50 mL of working
methyl red standard solutions. Also, 10 sample solutions were prepared with the quantity of its
components summarized in Table 1.1.

Determination of HMR and MR-, Molar Absorptivity Constant of HMR and MR-, and Acid
Dissociation Constant of HMR
The spectra of the previously prepared solution HMR and solution MR - between 350 and 600 nm were
obtained, having H2O in the reference cell. It was noted to use matched cells for all absorbance
readings. From these spectra, the wavelength of maximum absorption was determined for the acidic
and basic forms of the indicator, HMR and MR-, respectively. The absorbance of solutions 1-10 at HMR
and MR- was measured afterwards to determine the molar absorptivity constant of HMR and MR -, still
having H2O in the reference cell. To determine the acid dissociation constant of HMR, the absorbance
of samples 7-10 at both HMR and MR- was measured. Likewise, the pH of these solutions was also
measured using a pH meter.
