COLLECTION OF SPUTUM SPECIMEN

= a method wherein sputum is obtained for evaluation of gross appearance, for


microscope examination, for gram staining and culture, to identify the predominant organisms
and for cytologic examination.

PROCEDURE:

1. Secure early morning specimen. It yields the best sample of deep pulmonary material
from all lung fields.
2. Ask the patient to brush teeth or rinse mouth to decrease sputum contamination.
3. Instruct the patient to take several deep breaths, cough deeply and expectorate sputum
into a specimen container. The client should be encouraged to cough and not to spit so as
to obtain sputum.
4. See that the specimen is transported to laboratory immediately. Allowing the specimen to
stand in a warm room will result in overgrowth of organisms, making identification of
pathogen difficult. It also alters cell morphology.

COLLECTION OF URINE SPECIMEN

= a clean-catch midstream urine is the best clinically effective method of securing a


voided specimen for analysis so as to reduce as much as possible the contamination of the
specimen by external organisms.

PROCEDURE:

A. FOR MALE PATIENTS

1. Instruct the patient to expose glans and cleanse the area around meatus. Wash area with
mild antiseptic solution or liquid soap. Rinse thoroughly.
2. Allow the initial urinary flow to escape.
3. Collect the midstream urine specimen in a specimen container.
4. Avoid collecting the last few drops of urine because prostatic secretions may be
introduced into urine at the end of the urinary stream.
5. Send the specimen to the laboratory immediately.

B. FOR FEMALE PATIENTS

1. Ask the patient to separate her labia to expose the urethral orifice.
2. Cleanse the area around the urinary meatus with sponges soaked with antiseptic/soap
solution. Rinse thoroughly.
3. While the patient keeps the labia separated, instruct her to void forcibly.
4. Allow initial urinary flow to drain into bedpan or toilet, and then catch the midstream
specimen in a collection cup, making sure that the container does not come in contact
with the genitalia.
5. Send the specimen to the laboratory immediately.

COLLECTION OF STOOL SPECIMEN

= method of obtaining a stool specimen from the patient.
