Two-Dimensional NMR (2D NMR): COSY


correlation spectroscopy, COSY, is a pulse sequence used most frequently in 2D NMR. COSY is also the easiest 2D NMR method. It provides satisfactory spectra in three to four hours when several milligrams of the sample is available. With COSY almost all spin-spin couplings (including the long-range, which can complicate the analysis!) can be determined from a single chart. In COSY spectra: -Both vertical and horizontal axes show 1H shifts. -The 1D spectrum appears as "diagonal peaks". -Other signals (off-diagonal) are "cross peaks", which are due to 1H-1H spin coupling. To interpret a COSY spectrum: -First, draw a diagonal and distinguish the diagonal peaks from the cross peaks. -Then, draw horizontal and vertical lines from the cross peaks to find out which signals are coupled.

The cross peaks show that the CH2 and CH3 (H 1.27) protons are coupled, while one CH3 group (H 2.05) is not coupled to any hydrogen in the molecule.

The spectrum of F-ionone shows that H7 is coupled to H8. There are two long-range (4 bonds) couplings between H7 and 4-CH2 and 5-CH3 protons. The cross peaks are small since the couplings are weak. The coupling of 4-CH2 to 3-CH2, and that of 3-CH2 to 2-CH2 are clearly 6 observed.
