Batch Cultivation of Lactobacillus Sp.

for D-Lactic Acid Production Using Synthetic Whey Permeate as Production Medium and Optimization of Synthetic Whey Constituents by Plackett Burman and Response Surface Methodology (RSM).

D-Lactic acid (DLA) is one of desirable monomers to be used for the production of poly(D-lactic acid), PDLA, in bioplastics industry. PDLA is useful for improving thermostability of poly(L-lactic acid), PLLA, which is the main component of poly(lactic acid), PLA, biodegradable plastics. In this study, elite Lactobacilli strains viz., Lactobacillus delbrueckii subsp bulgaricus (LDB), Lactobacillus delbrueckii subsp lactis ATCC 4797(LDL1), Lactobacillus delbrueckii subsp lactis NCDO 2458 (LDL2) were employed for DLA production in Synthetic Whey Permeate medium (SWP medium). Lactobacilli strains were cultivated in SWP medium both at shake flask (pH 6.50.2, 37C and 150 RPM) and bench-scale bioreactor level (pH 6.5, 37C, 200 RPM and 20 % D.O). However, a controlled pH environment was ensured during batch cultivation of Lactobacilli strains in bioreactor experiments using intermittent dosing of acid/base and pH was maintained constant at 6.5. DLA final titer values were not significant in shake flask experiments, which were performed at un-controlled pH conditions (0.693 g/l for LDB and 2.5 g/l for LDL1). Moreover, another strain (LDL2) had not shown any significant production of DLA. Interestingly, bioreactor experiments indicated promising yield of DLA due to controlled pH environment especially LDL1 strain achieved a final maximum titer of 13.68 g/l for DLA within 23 h duration. LDL1 strain co-metabolized both lactose (100 % uptake) and casein (93 % uptake) effectively during its growth process, which substantiates its further application in fermentation of commercial whey permeate effluent for DLA production. Further media optimization was performed using 12 run Plackett Burman design to check effectiveness of Lactose, Casein, MgCl2, NaCl, KCl, CaCO3,Tween 80 and Ammonium citrate.
