Congratulations to Matthew R. Sullivan, et al. & Tania Konry from Northeastern University on their paper publication “Characterizing influence of rCHOP treatment on diffuse large B-cell lymphoma microenvironment through in vitro microfluidic spheroid model”! The study introduces a hydrogel spheroid-based microfluidic model for screening lymphoma treatments, aiming to address the challenges in improving patient survival rates for diffuse large B-cell lymphoma (DLBCL). Incorporating VitroGel® RGD into the hydrogel composition enhances cellular viability within the 3D culture environment. The RGD-rich hydrogel promotes cell adhesion, leading to increased viability of DLBCL and natural killer (NK) cells compared to alginate alone. Additionally, the rheological properties of the hydrogel more closely resemble those of metastatic lymph nodes, enhancing the model's physiological relevance. Congrats on Hermite® users' new Cell Press publication! A team from Shanghai University of Traditional Chinese Medicine showcases findings on the anti-tumor immunotherapy effects of the marine natural small molecule Benzosceptrin C. This molecule exerts its effects by targeting DHHC3 to induce lysosomal degradation of PD-L1. The authors utilized the Hermite® Uni-Fold functionality for predicting the 3D structure of DHHC3 and the Uni-IFD feature for the induced fit docking of all conformations of Benzosceptrin C with selected active sites. These computational approaches, combined with experimental validations, confirmed that Benzosceptrin C binds to the DHHC3 protein, inhibiting the expression of PD-L1 in tumor cells. Dr. Wang Jinxin, one of the article's authors, praised the simplicity and user-friendly interface of the tools used, highlighting the accuracy of the protein structure predictions. Particularly noteworthy was the induced fit docking results, where Uni-IFD was the only tool that accurately predicted and validated the binding modes observed. Lipid Nanoparticles Engineered to Target Lung Cells Reduce Tumor Size in Mice Lipid nanoparticles (LNPs) have enormous potential for the delivery of genomic medicine. One obstacle, however, is the lack of effective ways to target specific cell types. In order to target lung cells, the identification of chemically distinct lipid libraries would first be necessary. Now, a team of engineers has developed a new means of targeting the lungs with LNPs. More specifically, they report the implementation “of a barcoded high-throughput screening system as a means to identify the lung-targeting efficacy of cationic, degradable lipid-like materials.” The findings demonstrate a new method for efficiently determining which LNPs are likely to bind to the lungs, rather than the liver. This is published in Nature Communications in the paper, “High-throughput barcoding of nanoparticles identifies cationic, degradable lipid-like materials for mRNA delivery to the lungs in female preclinical models.” Typically, it would require hundreds of mice to individually test the members of a library of LNPs to find one with a low charge that has a higher likelihood of delivering a medicinal payload to the lungs. Instead, the team used barcoded DNA to tag each LNP with a unique strand of genetic material. More specifically, they combinatorially synthesized 180 cationic, degradable lipids. Then, they used barcoding technology to quantify how the selected 96 distinct lipid nanoparticles deliver DNA barcodes in vivo. After identifying an LNP that successfully penetrated lung cells—the top-performing nanoparticle formulation delivering Cas9-based genetic editors—they were administered to a lung tumor model in female mice. The treatment had a positive effect, drastically reducing tumor size by delivering a strand of mRNA and gRNA that suppresses the growth of lung tumors. Excited to share the most recent Cell Press publication from work carried out during my PhD in the Tomlinson lab, University of Leeds! 🧬 Here, high-content-imaging analysis combined with a wide-scale kinase RNAi screen, revealed PAK5 as a novel player in the progression of urothelial cancer. 🦠 The synthetic binding protein, Affimer 17 was subsequently raised against PAK5 using phage display technology. Intracellular expression of Affimer 17 was able to mimic the affects of PAK5 siRNA knockdown, through modulation of the protein conformation 🧫 🧪 Structural characterisation of the PAK5:Affimer17 complex unveiled a novel, intermediary conformation of kinase activation, providing a targetable chemical space for future drug discovery! 🔬 This work highlights the importance of using distinct biotechnologies to unlock novel, druggable conformations of not only kinases, but all major protein families! With their research, they showed that in patients with early BRCA mutations, the immune system fails to remove damaged breast cells, impairing the ability of the immune system to react at later stages. Single cell RNA-sequencing proved critical in their research. The approach complements and expands traditional RNA in situ hibridisation approaches, with the obvious limitations related to the availability of fresh tumor material.
