Congratulations to Anubhav Tripathi and team at Brown University for their recent publication focusing on a novel method for assessing dsRNA impurities that are generated during in vitro transcription of mRNA. In this article, they present a method that combines an enzyme, S1 nuclease, to identify and isolate dsRNA from an mRNA sample with a microfluidic electrophoresis analytical platform to characterize the impurity. After development and optimization, they tested their method using CATUG's CatPure™ dsRNA and mRNA samples. Results indicate that treatment led to complete disappearance of the main mRNA peak while the dsRNA fragments remained physically unaffected. The low sample volume and high throughput nature of the method makes it quite promising for batch-to-batch monitoring of residual dsRNA. However there are still some problems need to be concerned, such as the purity of mRNA substance. To ensure the property items of the quality control, arguments have lasted long in research and industrial fields. Finally, the USP guideline: Analytical Procedures for mRNA Vaccine Quality proclaimed in 2022 ended the chaos. 🔜 The mRNA USP guideline recommended several impurity items to be concerned, of which we noticed the residual T7 RNA polymerase. Residual enzymes in mRNA drug may cause immunogenicity, and furthermore rise the clinical risk. Excellent work. This again shows that drug disposition either via drug transporters or via drug metabolism is different in patients compared to human volunteers. These differences in patients can affect both efficacy and safety in patients in phase III trials. Preclinical Animal safety studies are conducted in healthy animals and Phase 1 trials are conducted in Healthy human volunteers who have no visible disease symptoms. We, at Reagene Biosciences, are developing 3D Non animal, Disease-in-A-Dish models to mimic human diseases in vitro and these models have translational value directly to patients and should be explored at pre clinical level. Such data can help eliminate animal use and phase 1 trials which have no direct relevance to patients. our ,3D models with simulated human disease can generate transcriptomic profiles those can be used as surrogate biomarkers for phase II and phase III clinical development.
