George Church and his team Harvard Medical School report the development and optimization of a platform for controlled, template-independent #enzymatic RNA #oligonucleotide synthesis. This enzymatic approach may address the length limitations commonly associated with phosphoramidite chemistry, as an enzymatic process has the potential for higher fidelity and fewer side reactions. Lipid nanoparticles (LNPs) are commonly used to deliver mRNA, and cationic lipids can significantly affect biodistribution, cellular uptake, endosomal escape, and transfection efficiency. However, cationic lipids are difficult to synthesize in the laboratory, which limits the discovery of suitable lipids and hinders large-scale production. In view of this, the researchers reported a one-pot series of multi-component reaction and designed a multi-component conjugation of amine-thiol-acrylate, which enabled the rapid synthesis (1 h) of degradable lipids containing amidines at room temperature. https://lnkd.in/g9eQxA8h
