UC San Diego Agilent Center of Excellence in Cellular Intelligence's weekly series--#DidYouKnow--features examples of how you can push the limits of #Measurements-#Across-#Scales. This is our second post of this series. #DidYouKnow that tissue #Scarring (#fibrosis) consumes #Energy? Activated #myofibroblasts exhibit significantly higher #energy consumption compared to their #quiescent state, due to increased #metabolic demands for #protein synthesis, #proliferation, and extracellular matrix production. By contrast, quiescent myofibroblasts maintain lower metabolic activity, conserving energy while preserving homeostasis. What was done here: •#1: Human colonic myofibroblasts isolated from an unaffected (normal) gut of patient enrolled at UC San Diego #IBD clinic were simultaneously assessed for Oxygen Consumption Rate (OCR), an established measure of mitochondrial function and extracellular acidification rate (ECAR), an indicator of glycolysis using #Seahorse XF Pro analyzer. ​•#2: Cells isolated from an affected segment of the gut of a patient with a #stricturing form of #CrohnsDisease (#CD) was assessed as above. Together, the two controls provide a HTP platform to screen for therapeutics that suppress the abnormal metabolic drive in myofibroblasts, which could halt/reverse intestinal #fibrosis, a devastating aspect of #stricturing #CD that has no #cure!
