Partnering with Advanced Research Projects Agency for Health (ARPA-H) for our VECTORS project grant is a significant milestone in realizing our vision for the future of gene therapies. While the benefits of expanding our capabilities and infrastructure are countless, here are four transformative ways this grant allows us to accelerate innovation. 1. Reducing Costs & Speeding Timelines: Through our work with ARPA-H, we will rapidly advance critical research initiatives, including refining our plant-based bioreactor technology to increase efficiency further and reduce production costs for AAV at 10x compared to current methods. 2. Scaling Production Capabilities: Expanding our production facility increases our capacity by more than 5x, allowing us to meet the growing demand for gene therapy products. This scale-up is essential to support large-scale manufacturing as we move toward commercialization. 3. Driving Strategic Collaboration: The grant resources provided through the grant open doors for partnerships with leading experts and organizations seeking innovative approaches in AAV development to launch new therapeutic programs at a lower cost and with shorter timelines. 4. Advancing Patient Access: Our ultimate mission is to improve global access to life-saving gene therapies. This grant allows us to significantly reduce the cost and time to deliver gene therapies to patients, making these treatments more affordable and accessible to those in need. Making AAV in a bioreactor is crazy expensive. But here’s how to make it in plants instead: (At a fraction of the cost) 1. Create plant-specific plasmids for encoding AAV components 2. Transform nanograms of the plasmids into plant bacteria 3. Infiltrate plants with bacteria 4. Bacteria releases plasmid, which kickstarts AAV production in the somatic cells of the leafs 5. Literally “juice” the plants once AAV production concludes (Up till now, none of this needs to be performed in a clean room environment - even when GMP batches are being made). 6. Transfer juice to GMP clean room 7. Perform multi-step clarification 8. Run through TFF filtration 9. Purify AAV with affinity chromatography, Iodixanol gradients or ion exchange chromatography 10. Formulate in your favorite excipient This is much less expensive. As in, 100x less. And the waste is biodegradable. If you’re attending Mesa next week… Let’s chat. This may look like an afternoon smoothie, but our "leaf blend" is a crucial part of our AAV production process. 🌿 In this step, AAV-producing leaves are homogenized in a blender and our biologic can be extracted from the leaves for downstream purification. Similar to tissue homogenization, the mechanical shear from the blades ruptures the plant cells and releases our AAVs into solution. #GeneTherapyManufacturing #PlantScience #ViralVector #Plants #AAV
