MTT assay

The MTT assay is a colorimetric assay for assessing cell metabolic activity. NAD(P)H-dependent cellular oxidoreductase enzymes may, under defined conditions, reflect the number of viable cells present. These enzymes are capable of reducing the tetrazolium dye MTT 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide to its insoluble formazan, which has a purple color. Other closely related tetrazolium dyes including XTT, MTS and the WSTs, are used in conjunction with the intermediate electron acceptor, 1-methoxy phenazine methosulfate (PMS). With WST-1, which is cell-impermeable, reduction occurs outside the cell via plasma membrane electron transport. Tetrazolium dye assays can also be used to measure cytotoxicity (loss of viable cells) or cytostatic activity (shift from proliferation to quiescence) of potential medicinal agents and toxic materials. MTT assays are usually done in the dark since the MTT reagent is sensitive to light.

MTT and related tetrazolium salts

MTT, a yellow tetrazole, is reduced to purple formazan in living cells. A solubilization solution (usually either dimethyl sulfoxide, an acidified ethanol solution, or a solution of the detergent sodium dodecyl sulfate in diluted hydrochloric acid) is added to dissolve the insoluble purple formazan product into a colored solution. The absorbance of this colored solution can be quantified by measuring at a certain wavelength (usually between 500 and 600 nm) by a spectrophotometer. The degree of light absorption depends on the solvent.

Track time trial

In the track time trial, a track cycling event, cyclists compete individually against the clock to record the fastest time over the specified distance from a standing start.

The bike

Track time trial bikes differ from normal track bikes in two major ways; firstly they have disc or 3–5 spoked wheels and secondly they often use aero-bars to allow the rider to adopt a more aerodynamic riding position, for the same reasons as in road individual time trials. The frames are often more streamlined to reduce air resistance.

Men – 1 km Time Trial

At the UCI Track World Championships, the distance for senior men is 1000 metres, hence the event's alternative name, the 'Kilo', short for kilometer. Junior men race over 500 m. Being such a short, high-intensity event, the kilo is popular with riders who specialise in the sprint. The Kilo time trial was removed from the Olympics programme after 2004 to make way for BMX racing. This led to a number of Kilo riders, most notably Chris Hoy, to begin focusing on other sprint events.

The AAT is a Trade Federation vehicle that appears in Star Wars Episode I: The Phantom Menace, Star Wars Episode III: Revenge of the Sith, the Star Wars expanded universe, and The Clone Wars. Early drafts of The Phantom Menace described the Trade Federation's invasion of Naboo being led by "armored attack craft" that resembled helicopters. BARC speeder

The BARC speeder appears in Revenge of the Sith and is a playable vehicle in Star Wars: Battlefront, Battlefront II and Star Wars Galaxies. They perform cavalry, scouting and mounted infantry roles and are armed with a single, or sometimes two, blasters. Inexpensive, lightweight, easy to maintain and able to fill various functions, they are a widespread mainstay of many armed organisations, governments and major armed forces.
