Ages Ago

Ages Ago is a musical entertainment with a libretto by W. S. Gilbert and music by Frederic Clay that premiered on 22 November 1869 at the Royal Gallery of Illustration. It marked the beginning of a seven-year collaboration between Gilbert and Clay. The piece was a critical and popular success and was revived many times, including at St. George's Hall, London in 1870 and 1874, and in New York in 1880.

Background

By the 1850s, the London stage had fallen into disrepute. Shakespeare's plays were staged, but most of the entertainments consisted of poorly translated French operettas, risque Victorian burlesques and vulgar broad farces. To bring family-friendly entertainment back to the theatre, Thomas German Reed and his wife Priscilla opened their Gallery of Illustration in 1855 and brought in Gilbert in 1869 as one of their many playwrights. The Gallery of Illustration was a 500-seat theatre with a small stage that only allowed for four or five characters with accompaniment by a piano, harmonium and sometimes a harp.

    The Argonaute protein family plays a central role in RNA silencing processes, as essential catalytic components of the RNA-induced silencing complex (RISC). RISC complex is responsible for the gene silencing phenomenon known as RNA interference (RNAi). Argonaute proteins bind different classes of small non-coding RNAs, including microRNAs (miRNAs), small interfering RNAs (siRNAs) and Piwi-interacting RNAs (piRNAs). Small RNAs guide Argonaute proteins to their specific targets through sequence complementarity (base pairing), which then leads to mRNA cleavage or translation inhibition.

    Discovery

    The RNA interference (RNAi) was first reported in 1995 by Guo and Kemphues, and similar pathways collectively referred to as RNA silencing were discovered in plants and fungi. The beginning of people’s understanding of the mechanism of RNA silencing began only in 1998 with the experiments of Fire and colleagues demonstrating that double-stranded RNA triggered RNAi. RNA silencing pathways process long RNAs into small RNAs that direct the repression of transcription or translation of nucleic acid targets with sequence corresponding to the small RNAs. These single-stranded RNAs, referred to as guide strands, are incorporated into RNA silencing effectors complexes such as the RNA-induced silencing complex (RISC). These RNA silencing effector complexes contain Argonaute family proteins.

    Ages is Tangerine Dream leader Edgar Froese's fourth studio album, released in 1978. Originally a double LP, it was reissued by Virgin Records in 1997 on one CD, with the closing track "Golgatha and the Circle Closes" missing. Froese has since remixed the album and released it on his own Eastgate label in 2005. This version also omitted a track, but not the same one as before, and another song, "Metropolis", was shortened.

    The album was recorded in Autumn 1977 at Amber Studios in Berlin. The drums and percussion were played by Klaus Krüger (aka Klaus Krieger) who was in Tangerine Dream at the time. Some parts of songs on the album were later reused on Tangerine Dream's albums, most notably "Nights of Automatic Women", which is very similar to "Madrigal Meridian" from Cyclone. Before the album was released, "Ode to Granny A" was released in an abridged form as the b-side to Tangerine Dream's live single "Encore", under the title "Hobo March".

    Reception

    On its initial release, the album failed to chart in any territory.

The Second Age extends to 3441 years, beginning with the foundation of Mithlond (the Grey Havens) under Círdan, and Lindon as the Noldorin Kingdom under Gil-galad, and ending with the defeat of Sauron at the hands of the Last Alliance of Elves and Men. The Third Age extends to 3021 years, ending with the final defeat of Sauron in the War of the Ring and the establishment of the Reunited Kingdom of Arnor and Gondor.. The Fourth Age is outside the scope of Tolkien's legendarium, beginning the suggested transition to the historical period, but Tolkien gives a summary of the first two centuries of the Fourth Age.
