The myeloblast is a unipotent stem cell, which will differentiate into one of the effectors of the Granulocyte series.

Origin [link]
These cells descend from the primitive reticulum cells, which are found in the stroma of the marrow. There is also an intermediate phase between the myeloblast and these primitive reticulum cell, namely the hemocytoblast. At this time several developing blood cell lines are available, like erythropoiesis and thrombopoiesis. The granulopoiesis is regulated by humoral agents, like Colony Stimulating Factor and Interleukin 3.
Location in the body [link]
The myeloblasts reside extravascularly in the marrow. The hematopoiesis takes place in the extravascular cavities between the sinuses of the marrow. The wall of the sinuses is composed with two different types of cells, the endothelial cells and the adventitial reticular cells. The hemopoietic cells are aligned in cords or wedges between these sinuses, the myeloblasts and other granular progenitors are concentrated in the subcortical regions of these hemopoietic cords.

Myeloblasts are rather small cells with a diameter between 14 and 18μm, from which the major part is occupied by a large oval nucleus. Their nucleus is composed of very fine nonaggregated chromatin and possesses 3 or more nucleoli. The cytoplasm has basophilic character and is devoid of granules, which is a major difference with its successor, the promyelocyte. The nucleolus is the site of assembly of ribosomal proteins, which are located in various particles dispersed over the cytoplasm. Mitochondria are present but have a rather small size.
The main features that distinguish a myeloblast from a lymphoblast upon microscopic examination are the presence of more prominent nucleoli, the nuclear chromatin being less condensed, and cytoplasmic granules are present.[1]

The granulopoiesis consists of 5 stages, of which the myeloblast is the first recognizable cell. Next in the differentiation sequence is the pro-myelocyte, this one will have ability to turn in one of the three different precursor cells, the neutrophilic, basophilic or eosinophilic myelocyte. This proliferation needs five divisions before the final stage is obtained. These divisions all take place in the first three stages of granulopoiesis.

Pathology [link]
Most common problem with malfunctioning myeloblasts is the acute myeloblastic leukemia. The main clinical features are caused by failure of the hemopoiesis with anemia, hemorrhage and infection as result. There is a progressive accumulation of leukemic cells, because some blast progenitor cells renew themselves and will have a limited differentiated division. Key mediators are the cell-density and Colony Stimulating Factor(CSF). A few acute myeloblastic leukemia can be initiated by earlier hematologic disorder, like pancytopenia, hypoplasia of the bone marrow.

Blast (Russian band)
Blast is a Moscow-based band formed in the late 1990s by Georgian singer/songwriter Nash Tavkhelidze.
He had previously spent a number of years in the US playing in different bands. The band was composed of Nash, Russian Alexandre "Khlap" Artchevski and Bulgarians Vlado Kostov and Valio Blagoev. At that time the Moscow club scene was booming and BLAST very quickly became the most popular indie band in the city. In 1998 band was picked up by the indie label "Apollo G Records" (Manchester, UK). They released their first album "Pigs Can Fly" and followed this with a UK club tour in 2000. A few years later the band signed to Ghost Records UK and hit the studio recording the album F**K the industry with producer Graham Pilgrim. Ghost Records Musical Director Frank Perri and A&R manager Phillipe Palmer drew much attention to the Russian Rock Rebels and a place within the British music scene was forged. Regular tours of the UK and Europe have continued since that time.

23 Blast is a 2013 American sports drama film directed by Dylan Baker. The film was written by Bram and Toni Hoover, inspired by the story of Travis Freeman, a Kentucky teen who loses his sight, but eventually overcomes the challenges of his disability, and continues to live his dream of playing football. Travis is portrayed by Mark Hapka in the film.
The film was produced by Touchdown Productions, LLC and Toy Gun Films. Stephen Lang, Alexa PenaVega,Max Adler and Becky Ann Baker also starred in the film.
Plot
Travis Freeman, a local football star, is an average high school junior. Living in the town of Corbin, Kentucky, his inner circle includes his supporting parents, girlfriend, and childhood best friend, Jerry Baker (Bram Hoover) Life is normal for Travis until one day he is stricken with a series of severe headaches and a bacterial meningitis infection. After being rushed to the hospital, Travis undergoes surgery to remove the infection and save his life. He comes out of the procedure alive, but he is now blind. After losing his sight, Travis experiences depression as he adjusts to life without sight. When forced to abandon football, Travis’ shallow, cheerleader girlfriend quits him, and he faces the decision of having to attend the school for the blind away from home. With the help of his parents, best friend, Jerry, another childhood friend, Ashley, and his mobility coach, Travis pulls through and is able to adapt to his new disability and starts his senior year at Corbin High School. Every day after school, Travis attends football practice, but watches, with help from his friend, Ashley, from the sidelines. His coach and mentor, Coach Farris, approaches him one day about joining the team again. Without much convincing, Travis rejoins the football team as a center and helps the team turn around their losing season advancing to the state playoffs.

In statistics, the so-called 68–95–99.7 rule is a shorthand used to remember the percentage of values that lie within a band around the mean in a normal distribution with a width of one, two and three standard deviations, respectively; more accurately, 68.27%, 95.45% and 99.73% of the values lie within one, two and three standard deviations of the mean, respectively. In the empirical sciences the so-called three-sigma rule of thumb expresses a conventional heuristic that "nearly all" values are taken to lie within three standard deviations of the mean, i.e. that it is empirically useful to treat 99.7% probability as "near certainty". The usefulness of this heuristic of course depends significantly on the question under consideration, and there are other conventions, e.g. in the social sciences a result may be considered "significant" if its confidence level is of the order of a two-sigma effect (95%), while in particle physics, there is a convention of a five-sigma effect (99.99994% confidence) being required to qualify as a "discovery".
Sigma factor
A sigma factor (σ factor) is a protein needed only for initiation of RNA synthesis. It is a bacterial transcription initiation factor that enables specific binding of RNA polymerase to gene promoters. The specific sigma factor used to initiate transcription of a given gene will vary, depending on the gene and on the environmental signals needed to initiate transcription of that gene.
Every molecule of RNA polymerase holoenzyme contains exactly one sigma factor subunit, which in the model bacterium Escherichia coli is one of those listed below. The number of sigma factors varies between bacterial species.E. coli has seven sigma factors. Sigma factors are distinguished by their characteristic molecular weights. For example, σ70 refers to the sigma factor with a molecular weight of 70 kDa.
RNA polymerase holoenzyme complex consisting of core RNA polymerase and a sigma factor executes transcription of a DNA template strand. Once initiation of RNA transcription is complete, the sigma factor can leave the complex.
