Known for most of its length as Kearneysville Pike, the state highway runs {{convert|5.6|mi|km}} from [[West Virginia Route 115|WV 115]] in [[Kearneysville, West Virginia|Kearneysville]] north to the [[Maryland]] state line at the [[Potomac River]] in [[Shepherdstown, West Virginia|Shepherdstown]], where the highway continues as [[Maryland Route 34]] (MD 34). WV 480 is one of the main north–south highways of northern [[Jefferson County, West Virginia|Jefferson County]] and passes through the campus of [[Shepherd University]].
WV 480 was formerly WV 48 before [[U.S. Route 48 (Maryland-West Virginia)|US 48]] (now [[I-68 (WV)|I-68]]) entered the state.
